[摘要]
目的: 通过比较NaNO2-Al( NO3)3-NaOH 法和AlCl3法的差异,建立荞麦降糖颗粒中总黄酮含量的
比色分析方法。方法: 以乙醇为溶剂进行回流提取,并以芦丁为对照品,分别以NaNO2 -Al( NO3)3 -NaOH 和
AlCl3为显色剂,分别在516nm 和417nm 处用可见分光光度法测定荞麦降糖颗粒中总黄酮含量。结果: 建立硝酸
铝法芦丁浓度-吸光度关系的回归方程方程式y = 0.01201x-0.0291,R2 = 0.9994,回收率为100.2%。同时也建立
三氯化铝法芦丁浓度-吸光度关系的回归方程方程y = 0.02992+0.00473,R2 = 0.9996,回收率为99.75%。结论:
AlCl3法操作简便快速、专属性强,更适用于荞麦降糖颗粒中总黄酮的含量测定。

[Abstract]
Objective: By comparing the difference between the NaNO2 - Al ( NO3)3 - NaOH
methods and AlCl3methods, to set up a Colorimetric assay for the determination of total flavones in the
Buckwheat Hypoglycemic granule.Methods: Extract the total flavones by ethanol,with rutin as standard
sample,NaNO2-Al( NO3)3-NaOH and AlCl3 as color reagents, the flavonoids content was determined
by VlS-spectrophotometry at the wavelength of 516nm and 417nm.Result: The regression equation of
NaNO2-Al( NO3)3-NaOH methods was y = 0.01201x-0.0291,R2 = 0.9994, the average recovery of this
method was 100.2%. The regression equation of AlCl3 methods was y = 0.02992 x + 0.00473,R2 =
0.9996, the average recovery of this method was 99.75%.Conclusion: The method of spectmphotometry
in the AlCl3 is simple, rapid,highly accurate,and suitable for quality control of Buckwheat Hypoglycemic
granule.
