目的: 探讨胰腺癌细胞Wnt 通路中核心分子β－catenin 的mRNA 表达水平与其化疗耐药的相关性。
方法: 使用Real－time PCR及Taqman 探针法检测其β－catenin mRNA 表达水平，胰腺癌细胞株AsPC－1，MIAPaCa
－2，T3M4，BxPC－3 与COLO357 的化疗耐药检测使用CCK－8 试剂盒。结果: Wnt 通路中β－catenin 在不同类型的
胰腺癌细胞中表达mRNA 水平差异明显，细胞株T3M4，AsPC－1 与MIAPaCa－2 中β－catenin mRNA 水平低( 低β
－catenin 表达组) ，而COLO357 与BxPC－3 中β－catenin mRNA 表达水平较高( 高β－catenin 表达组) ，胰腺癌细胞
T3M4，AsPC－1 与MIAPaCa－2 作为β－catenin 低mRNA 表达组的5－FU 抑制率3 个差异药物浓度高中低各自是
( 0．891±0．076) 、( 0．812±0．098) 和( 0．724±0．093) 。相同化疗浓度下胰腺癌细胞COLO357 与BxPC－3 作为β－
catenin 高mRNA 表达组对5－FU 抑制率分别是( 0．625± 0．038) 、( 0．536± 0．033) 和( 0．368± 0．057) 。β－catenin
mRNA 表达低的细胞对5－FU 化疗敏感性明显高于β－catenin 表达高的细胞，差异有统计学意义( P＜0．05) 。结
论: 胰腺癌细胞5－FU 的化疗敏感性与β－catenin 的mＲNA 表达水平对关系紧密。

[Abstract]
Objective: To investigate the relationship expression of beta－catenin mRNA and chemoresistance
in pancreatic cancer cells to 5－FU．Methods: The chemosensitivity of five pancreatic carcinoma
cell lines to 5－FU in vitro was evaluated with CCK－8 kit．β－catenin mRNA expression of pancreatic
carcinoma cells was analyzed by Real－time PCR.Results: Pancreatic carcinoma cells T3M4，As-
PC－1，MIAPaCa－2 expressed low amounts of β－catenin mRNA( Low β－catenin group) ．Cells of COLO357
and BxPC－3 expressed high amounts of β－catenin mRNA( High β－catenin group) in five cell
lines．The inhibition rates( IRs) of Low β－catenin group cells for high，middle and low dose to 5－FU
were( 0．891±0．076) ，( 0．812±0．098) and( 0．724±0．093) respectively．The IRs of High β－catenin group
for high，middle and low dose to 5－FU were( 0．625±0．038) ，( 0．536±0．033) and( 0．368±0．057) respectively．
IRs of 5－FU tended to be lower in the cells with high β－catenin mRNA expression than
those with low β－catenin mRNA expression．The difference reached statistical significance( P＜0．05) ．
Conclusion: β－catenin mRNA expression levels were significantly correlated to 5－FU sensitivity in five
pancreatic carcinoma cell lines．
