[摘要]
目的: 探讨TNF-α 对HCAEC 的NF-kB mRNA 的影响,探讨人冠状动脉内皮细胞内的炎症过程的
分子机制。方法: 采用人冠状动脉内皮细胞培养传代后作为实验对象,随机分为空白组和诱导组,分2 h、6 h、10
h 三个时间点,每个时间点以2.5μg /L、5μg /L、10μg /L 的浓度刺激HCAEC 后,提取细胞RNA 并逆转录,用RTqPCR
法检测NF-kB mRNA 的转录水平。结果: TNF-α 可抑制HCAEC 的NF-kB mRNA 转录水平( P<0.05) 。其
中NF-kB 5μg /L,2 h、10μg /L,2 h、10μg /L,6 h 诱导组mRNA 含量较正常对照组有显著差异( P<0.05) 。且NFkB
mRNA 的浓度变化与TNF-α 的刺激浓度和时间有一定的趋势规律。结论: TNF-ɑ 在一定时间范围内对
HCAEC μg /L 的mRNA 的转录和蛋白质的表达水平具有抑制作用。

[Abstract]
Objective: To investigate the correlation between the TNF-ɑ and Nuclear factor———kB
NF-kB mRNA expression of human coronary artery endothelial cells( HCAEC) , to explore the part of
the pathogenesis of inflammation in HCAEC.Methods: CμLtured coronary artery endothelial cells represented
as experimental objects,were randomly divided into blank group and the inducted group, induced
group was divided into blank group, induction group including 2h,6h,10h three time points,
each time point had 2.5μg /L,5μg /L,10μg /L induced group.After stimμLation to HCAEC,cellμLar
RNA extraction and reverse transcription followed.Transcriptional level of NF-kB m’RNA was detected
by RT-qPCR.Results: TNF-α might in a certain time frame inhibit NF-kB m’RNA transcription level
of HCAEC( P < 0.05) .Wherein the NF - kB 5μg /L,2 h、10μg /L,2 h、10μg /L,6 h group induced
mRNA levels were significantly different( P< 0.05) compared with normal control group.And NF -kB
mRNA changed with TNF-α stimμLation and concentration by a certain trend regμLarity.Conclusion:
The expression of mRNA transcription of NF-kB were inhibited by TNF-α in human coronary artery
endothelial cells.
