[摘要]
目的: 通过研究11β 羟基类固醇脱氢酶2( 11β-Hydroxysteroid dehydrogenase Type 2,11β-HSD2) 与
肿瘤坏死因子-α( tumor necrosis factor-alpha,TNF-α) 在妊娠期高血压疾病病人胎盘组织中的表达及相互关系,
探讨两者在该病的发生、发展中的作用及意义。方法: 选取孕妇共120 例,其中正常妊娠组40 例,妊娠期高血压
疾病组共80 例( 其中子痫前期轻度组40 例,子痫前期重度组40 例) 。上述病人均于胎盘娩出后取胎盘母体面
组织块,通过实时荧光定量PCR 法检测三组病人胎盘组织中11β-HSD2、TNF-α 的mRNA 的表达,并分析两者
间的相关性。结果: ( 1) 妊娠期高血压疾病组的11β-HSD2 mRNA 表达水平均明显低于正常妊娠组,且子痫前期
重度组的表达水平明显低于子痫前期轻度组( P<0.05) 。( 2) 妊娠期高血压疾病组TNF-αmRNA 的表达水平均
高于正常妊娠组,且子痫前期重度组的表达明显高于子痫前期轻度组( P<0.05) 。( 3) 在妊娠期高血压疾病病人
胎盘组织中11β-HSD2与TNF-α 的表达存在负相关关系( r = -0.467,P<0.05) 。结论: TNF-α 可能参与了11β-
HSD2的调节,并抑制其活性,二者可能通过一些直接或间接的作用,共同参与了妊娠期高血压疾病的发生、
发展。

[Abstract]
Objective: To detect the expression and relationship of 11β-HSD2 and TNF-α in placenta
with hypertension disorders complicating pregnancy.In order to explore the effect and significance
of the occurrence and progression of this disease. Methods: 120 pregnant women were selected,
including 40 patients of normal pregnancy group,80 cases of hypertensive disorders complicating pregnancy
group( including 40 patients of preeclampsia in mild group,40 patients of severe preeclampsia
group) .I took their placental tissue,and detected the expression of 11β-HSD2 and TNF-α in three
groups by Real-Time PCR method.The correlation between 11β-HSD2 and TNF-α was evaluated.
Results: ( 1) The expression level of 11β-HSD2 mRNA in hypertensive disorders complicating pregnancy
group is significantly lower than that in the normal pregnancy group.And that in severe preeclampsia
group is significantly lower than that in preeclampsia mild group( P<0.05) .( 2) The expression level of
TNF-α mRNA in hypertensive disorders complicating pregnancy group is significantly higher than that
in the normal pregnancy group.And that in severe preeclampsia group is higher than that in preeclampsia
mild group( P<0.05) .( 3) The expression levels of 11β-HSD2 and TNF-α in hypertensive disorders
in pregnancy are negative correlation( r = -0.467,P<0.05) .Conclusion: TNF-α may be involved in the
regulation of 11β-HSD2,and inhibit its activity.They probably interact with each other through some
direct or indirect way.They both participate in the development of hypertensive disorders in pregnancy.
