[摘要]
目的: 探讨BRCA-1、PARP-1 的表达变化与乳腺组织、三阴乳腺癌及非三阴乳腺癌的关系,探讨其
在三阴乳腺癌治疗、预后评估中的预测价值,以期望PARP-1 及BRCA-1 成为新的预测影响因子,为治疗靶点提
供理论依据。方法: 收集我院2015-08~ 2016-08 期间病人的新鲜乳腺组织共90 例。采用RT-PCR 技术进行分
析,分别研究BRCA-1、PARP-1 在不同乳腺组织表达差异,说明BRCA-1、PARP-1 在三阴乳腺癌发生、发展中
的作用。结果: ( 1) 三阴乳腺癌的BRCA- 1 的表达低于非三阴乳腺癌( t = 3.202,P< 0.001) ; 乳腺恶性肿瘤的
BRCA-1 的表达低于乳腺良性肿瘤( t = 9.335,P<0.001) ; 且BRCA-1 在乳腺肿瘤中的表达,不论良恶性,都低于
正常乳腺组织( t = 18.813,P< 0.001) 。( 2) 三阴乳腺癌的PARP - 1 的表达高于非三阴乳腺癌( t = 5.622,P<
0.001) ; 乳腺恶性肿瘤的PARP-1 的表达高于乳腺良性肿瘤( t = 10.673,P<0.001) ; 且PARP-1 在乳腺肿瘤中的
表达,不论良恶性,都高于正常乳腺组织( t = 10.016,P<0.001) 。( 3) 随着BRCA-1 表达值的增高,PARP-1 的表
达呈下降趋势,两者呈负相关( r = -0.817,P<0.001) 。结论: BRCA-1 参与乳腺癌的发生发展,随着肿瘤恶性程度
的增加,BRCA-1 的表达呈下降趋势,在三阴乳腺癌中的表达高于非三阴乳腺癌,随着肿瘤恶性程度的增加,
PARP-1 的表达呈上升趋势,在三阴乳腺癌中的表达高于非三阴乳腺癌。

[Abstract]
Objective: To analyze the predictive value of BRCA-1 and PARP-1 expression in normal
breast tissues, triple negative breast cancer and non-triple negative breast cancer,and discusses its
treatment and prognostic evaluation of predictive value in triple negative breast cancer, in order to expect
PARP-1 and BRCA-1 to become the new predictive factors,which will provide theoretical basis
for the therapeutic target.Methods: Collecting 90 cases of fresh breast tissue we used RT - PCR to
measure BRCA-1 and PARP-1 expression levels, to investigate the difference of expression of BRCA-
1 and PARP-1 in different breast tissues,and to analyze the correlation between BRCA-1 and PARP-
1 and breast cancer patients’age,sex, tumor diameter,depth of invasion,degree of differentiation,
lymph node metastasis,TNM stage and follow-ups after operation and its prognosis.Results: ( 1) The
expression of BRCA-1 in the TNBC is lower than that in the non-TNBC( t = 3.202,P<0.001) has statistical
significance.The expression of BRCA-1 in the breast tumors is lower than that in the normal
breast tissues( t = 18.813,P<0.001) ( 2) The expression of PARP-1 in the TNBC is lower than that in
the non -TNBC( t = 3.202,P < 0.001) has statistical significance. The expression of PARP - 1 in the
breast tumors is higher than that in the normal breast tissues( t = 10.016,P<0.001) ( 3) With the increasing
of the expression of the BRCA-1, the expression of the PARP -1 is decreasing, so the both
ways are negatively correlated with statistical significance( r = -0.817,P<0.001) .Conclusion: With the
development of breast cancer, the expression of the BRCA- 1 is decreasing with the increase of the
tumors’ malignant degree.The expression of the BRCA-1 in triple negative breast cancer is higher than
that in non -triple negative breast cancer,which will become a new target for screening therapeutic
method.Nevertheless, the expression of the PARP-1 is increasing with the rising of the tumors’ malignant
degree involved in the development of breast cancer.The expression of the PARP-1 in triple negative
breast cancer is higher than that in non-triple negative breast cancer, the PARP-1 inhibitor may
benefit from the treatment of TNBC as a target of inhibitor.
