[摘要]
目的: 基于网络药理学探讨红花的活性药效物质基础,并分析其多种药理作用机制。方法: 在中药 系统药理数据库和分析平台( TCMSP) 中搜索红花的化学成分,结合口服生物利用度及“类药五原则”筛选出红 花的潜在活性成分,通过在线靶点预测平台和 CTD 数据库收集靶点与疾病相关信息,应用 DAVID 生物信息学资 源数据库富集关键靶点相关的主要生物通路,采用 Cytoscape 软件,构建红花“成分-靶点”、“靶点-通路”、“成分 -疾病”等互作网络图,进而分析红花的多种药理作用机制。结果: 共获得黄酮类、生物碱类、木脂素类等化合物 475 个,经筛选后最终确定 25 个主要活性成分; 作用于靶点 318 个,其关键靶点有 TP53、AKT 1、ESR 1 等,涉及 显著性通路 59 条,其中与肿瘤、神经系统疾病及心血管等疾病相关程度较高。结论: 通过网络药理学成功预测 红花的活性成分及作用机制,为其深层次的开发与利用奠定基础。

[Abstract]
Objective: To investigate the active pharmacodynamic basis of Safflower and analyze its multi - dimensional pharmacological mechanism based on network pharmacology. Methods: The chemical components of safflower were searched in the TCMSP database,combining the oral bioavail- ability and ’five principles’ as the screening conditions to obtain active compounds.The potential targets and diseases information were collected through the online target prediction platform and CTD online a- nalysis platform.The key biological pathways associated with key targets were enriched by using DAVID database.Constructing the component-target,target-pathway and component-disease network by Cyto- scape software to analyze the various pharmacological mechanisms of safflor. Results: A total of 475 compounds were obtained by systematically collecting safflower chemical constituents,such as fla- vonoids,alkaloids,and lignans. 25 components were finally determined after screening; These compounds acted on 318 targets,the key targets were TP53,AKT 1,and ESR 1,etc.Involving 59 signif- icant pathways are highly related to the diseases such as tumors,nervous system diseases and cardiovas- cular diseases.Conclusion: The active components and mechanism of Safflower were successfully pre- dicted by network pharmacology,which established the foundation for its deep development and utilization.
