: 目的: 针对 2 型糖尿病( T2DM) 患者单核细胞之中糖类反应组件结合蛋白( ChREBP) 进行分析,确 定其基因启动子范围的甲基化程度,并分析其与 T2DM 之间的相关性。方法: 空腹抽取研究目标的静脉血液,肝 素进行处理,分析测试机检测其在空腹状态下的生化指标,通过针对甲基化的 PCR 亚硫酸氢盐限制性内切酶法 ( BSP) 考察 ChREBP 的启动子甲基化程度均值。结果: T2DM 组病人 ChREBP 基因启动子的甲基化程度均值在 P<0.05的条件下较对照组更高; 对于实验组,其 ChREBP 基因之中的 8 个 CpG 位点和对照组相比拥有更高的甲 基化程度,且在 P<0.01的条件下存在统计学上的显著差别。结论: 2 型糖尿病病人具有更高的 ChREBP 基因启 动子甲基化程度,所以本研究针对 ChREBP 基因进行分析,考察其启动子 CpG 位点的甲基化程度,结果显示,该 甲基化程度的不正常可能和糖尿病存在一定的相关性。

[Abstract]
Objective: For the type 2 diabetes mellitus( T2DM) patients,the carbohydrate reaction component binding protein( ChREBP) was analyzed to determine the methylation degree of the gene promoter range,and analyzed with T2DM.Correlation between.Methods: Under fasting conditions Tak- ing,the venous blood of the study target and treating with heparin to test the biochemical indicators, through methylation PCR bisulfite restriction endonuclease ( BSP ) was used to investigate the mean methylation degree of ChREBP promoter.Results: The mean methylation degree of ChREBP gene pro- moter in T2DM group was higher than that of the control group under P<0.05.For the experimental group,Eight of the CREG sites in the ChREBP gene had a higher degree of methylation than the control group,and there was a statistically significant difference at P<0.01.Conclusion: Patients with type 2 diabetes have a higher degree of methylation of the promoter of ChREBP gene. Therefore,this study aimed to analyze the ChREBP gene and investigate the promoter CpG site.The degree of methylation showed that the abnormal degree of methylation may be related to diabetes.
