[摘要]
目的 探讨轻中度肝纤维化大鼠肝脏表观扩散系数(apparent diffusion coefficient,ADC)与基质金属蛋白酶-1(matrixmetalloproteinase-1,MMP-1) 表达的相关性。方法 采用猪血清腹腔注射建立肝纤维化大鼠模型。采用完全随机设计、随机数字表抽样法,于造模第5周开始,第5-12周每周随机抽取实验组大鼠4只,对照组大鼠1只行DWI检查,共32只实验组大鼠及8只对照组大鼠行3.0 T MR DWI检查,b值选取800s/mm2,计算ADC值。MR检查后处死大鼠灌注固定,行HE、Masson三色染色、网状纤维染色,RT-PCR检测MMP-1 mRNA表达。统计学采用单因素方差分析、SNK法多重比较以及Pearson相关分析。结果 32只模型组大鼠中,其中纳入实验组轻度肝纤维化(S1期)11只、中度肝纤维化(S2期7只、S3期8只)15只;其余肝纤维化S0期3只、S4期1只,图像不符合分析要求2只。对照组、轻、中度肝纤维化组大鼠肝脏ADC值及MMP-1 mRNA表达差异有统计学意义(F值分别为38.66和41.72,P值均<0.05),对照组与轻度肝纤维化组ADC值及MMP-1 mRNA表达无统计学差异(P值>0.05),中度肝纤维化组与对照组及轻度肝纤维化组比较均有统计学差异(P值<0.05)。秩相关分析显示ADC值与MMP-1 mRNA表达呈正相关(r=0.64,P<0.05)。结论 随着肝纤维化进程大鼠肝脏ADC值及MMP-1 mRNA表达均逐渐减低,二者呈正相关。

[Abstract]
Objective To investigate the correlation between the ADC and expression of TIMP-1 mRNA in Mild to moderate liver fibrosis of rats. Methods Rat model with liver fibrosis was established by porcine serum intraperitoneal injection. By using completely randomized design random digits table, 4 rats were assigned to model group and 1 rat to the control group. After drug administration from the 5th week to the end of 12th weeks,a total of 32 rats of model group and 8 rats of control group were included to undergo DWI scanning. the value of ADC for b values=800 s/mm2 was calculated. The rats were sacrificed to death to have pathologic examination after DWI scanning immediately. By using real time-PCR to detect the expression of MMP-1 mRNA. Statistics analysis using one-way ANOVA, SNK multiple comparison and Pearson correlation. Results In the model group, Conclude Mild liver fibrosis(Grade S1) 11 rats, moderate liver fibrosis(Grade S2,S3) rats 15, there were 3 rats in grade 0, 1 in grade 4 of liver fibrosis, there were 2 rats do not accord with the requirements of the analysis in DWI scanning. The value of ADC and the expression of MMP-1 mRNA were significant differences among control group and Mild to moderate liver fibrosis groups(F=38.66 and 41.72,P<0.05). There were significant differences of the value of ADC and MMP- mRNA between Mild and control groups to moderate group(P<0.05). There was a positive correlation between the expression of TIMP-1 mRNA and ADC value(r=0.64. P<0.05). Conclusions As the value of ADC decreased in the progression of rats’ liver fibrosis , the expression of mRNA in MMP-1 decreased too, the two were positively correlated.
