目的:本研究探讨结肠癌组织中VM、EMT和Wnt通路关键蛋白表达的相关性。方法:选取200例结肠癌患者的组织标本,利用免疫组化测定VM、EMT的两种标记蛋白(Vimentin、E-cadherin)及Wnt通路调节蛋白β-catenin的表达,并探讨其相关性。结果:200例人结肠癌组织中有45例VM表达阳性,VM阳性患者肿瘤分化更差、分期更晚,更易发生复发转移,差异有统计学意义(P<0.05)。VM阳性时Vimentin高表达(66.7%),E-cadherin低表达(15.5%),差异有统计学意义(P<0.05)。同时VM阳性β-catenin为高表达(60.0%),VM阴性β-catenin为低表达(8.4%),差异具有统计学意义(P<0.05) 。β-catenin阳性的结肠癌组织中,Vimentin为高表达(72.5%),而E-cadherin低表达(30.0%),差异具有统计学意义(P<0.05)。结论:在结肠癌组织内存在VM现象,VM与EMT存在相关性。VM与Wnt通路存在相关性。结肠癌中Wnt通路活化可能促进EMT发生。

Abstract: Objective: To explore the correlation between vasculogenic mimicry (VM), epithelial-mesenchymal transition (EMT) and Wnt signaling pathway in colon cancer. Methods: Tumor tissues were collected from 200 colon cancer patients. Immunohistochemistry was performed to detect β-catenin, Vimentin and E-cadherin expression. The correlations of VM with β-catenin and EMT were analyzed. Results: Among 200 cases of human colon cancer, 45 cases had positive VM expression, VM was associated with tumor size and clinical stage (P < 0.01),and VM positive patients had worse tumor differentiation, with higher recurrence and metastasis rate (P < 0.05). VM positive associated with higher expression rate of Vimentin (66.7%) and lower expression rate of E-cadherin (15.5%), and the difference was statistically significant (P< 0.05). Meanwhile, VM positive associated with higher β-catenin expression (60.0%), and VM negative patients had lower expression rate of β-catenin (8.4%), the difference was statistically significant (P< 0.05). In colon cancer tissues with positive β-catenin, Vimentin was highly expressed (72.5%) and E-cadherin was low (30.0%), the difference was statistically significant (P< 0.05). Conclusion: VM formation in colon cancer, and VM is correlated with EMT. VM is associated with the Wnt signaling pathway. The activation of Wnt pathway in colon cancer may promote EMT in colon cancer.
