[摘要]
目的:探讨蒙药古日古木-13含药血清对H2O2诱导Huh-7细胞氧化应激损伤的保护作用及其可能机制。方法:40只大鼠随机分5组,分为古日古木-13高、中、低剂量组、阳性对照组及PBS空白组,灌胃7 d,于最后一次给药1 h后取血,制备含药血清培养基;CCK8法筛选H2O2损伤Huh-7细胞最适浓度及古日古木-13含药血清最适剂量浓度和作用时间;HPLC法检测古13含药血清中有效成分;流式细胞仪测细胞凋亡变化;qPCR法检测细胞NF-κB的表达变化;蛋白印迹法检测细胞NF-κB蛋白表达变化。结果:Huh-7细胞半数抑制率为930 mmol/ml;选择中剂量浓度古13含药血清,作为后续作用条件(P<0.05);H2O2+10%古13血清组作用Huh-7肝细胞的最适时间为18 h;古13含药血清的有效成分是没食子酸及鞣花酸;H2O2+10%古13血清组能降低H2O2损伤的Huh-7细胞凋亡率,差异有统计学意义(P<0.05);H2O2+10%古13血清组细胞NF-κB较H2O2+10%空白血清组细胞表达量降低,差异有统计学意义(P<0.05);H2O2+10%古13血清组细胞的NF-κB蛋白表达量明显低于H2O2+10%空白血清组蛋白表达量,差异有统计学意义,P<0.05。结论:蒙药古日古木-13含药血清对 H2O2 损伤的 Huh-7 细胞具有保护作用,其机制可能与NF-κB通路有关。

[Abstract]
Objective: To investigate the protective effect of Mongolian medicine Gurigumu-13 on oxidative stress injury of Huh-7 cells induced by H2O2 and its possible mechanism. Methods: 40 SD rats were randomly divided into three groups and treated with high, middle and low dose of Gulimu-13, tiopronin enteric-coated tablets and the same amount of PBS, respectively. After intragastric administration for 7 days, the blood was taken 1 hour after the last administration, and the drug-containing serum medium was prepared; the model concentration of Huh-7 cells damaged by H2O2 and the optimal dose concentration and action time of Gurigumu-13 serum were screened by CCK8 method; the effective components in Gu 13 serum were detected by HPLC method; the changes of apoptosis were detected by flow cytometry; the expression of NF-κB was detected by qPCR method; the expression of NF-κB protein was detected by Western blotting. Results: the 50% inhibition rate of Huh-7 cells was 930 mmol/ml;, the medium dose of gu 13 containing serum was selected as the follow-up condition, the optimum time for Huh-7 hepatocytes to be treated with H2O2+ 10% gu 13 serum was 18 h, and the effective components of gu 13 containing serum were Gallic acid and ellagic acid. The apoptosis rate of Huh-7 cells damaged by H2O2 was significantly decreased in H2O2 + 10% Gu 13 serum group, and the expression of NF-κB in H2O2 + 10% Gu 13 serum group was significantly lower than that in H2O2 10% blank serum group (P<0.05). The apoptosis rate of NF-κB in H2O2 + 10% Gu 13 serum group was significantly lower than that in H2O2 10% Gu 13 serum group (P < 0.05). The expression of NF-κB protein in H2O2 10% ancient 13 serum group was significantly lower than that in H2O2 10% blank serum histone group, and the difference was statistically significant. Conclusion: the serum containing Mongolian medicine Gurigumu-13 has a protective effect on Huh-7 cells injured by H2O2, and its mechanism may be related to NF-κB pathway.
