摘要:目的:建立RP-HPLC方法同时测定苓桂术甘汤中甘草苷和原儿茶酸的含量。方法:采用Eclipse Plus C18色谱柱,在260nm波长检测,柱温25℃,流速1.0mL· min﹣1,梯度洗脱,流动相为乙腈-0.1%磷酸水,进样量为10μL。结果:苓桂术甘汤中甘草苷和原儿茶酸的线性范围分别为2.12~21.15μg·mL-1(r=0.9992)、1.36~13.60μg·mL-1(r=0.9993);两成分的加样回收率的均值分别为98.37% (RSD=0.9%,n=6)、98.51%(RSD=1.1%,n=6);在此条件下,苓桂术甘汤中甘草苷和原儿茶酸的含量分别为618.6μg·g-1、49.09μg·g-1,RSD均为0.9%。结论:所用方法简单灵敏、快速高效、易于操作,可用于测定量苓桂术甘汤中甘草苷和原儿茶酸的含量。

Abstract: Objective: To establish a high performance liquid chromatography method for the simultaneous determination of glycyrrhizin and protocatechuic acid in Linggui Zhugan decoction. Methods: The chromatographic column was Eclipse Plus C18; the mobile phase was acetonitrile-0.1% phosphoric acid aqueous solution with gradient elution; the detection wavelength was 260nm; the flow rate was 1.0mL·min﹣1; the column temperature was 25℃. Results: There was a good linear relationship between glycyrrhizin and protocatechuic acid in Linggui Zhugan Decoction in the range of 2.12~21.15μg·mL-1,1.36~13.60μg·mL-1 . The correlation coefficients r were 0.9992 and 0.9993 , respectively. The average recovery of glycyrrhizin was 98.37% (RSD=0.9%, n=6) , and the average recovery of protocatechuic acid was 98.51% (RSD=1.1%, n=6) .Under these conditions, the contents of glycyrrhizin and protocatechuic acid in Lingguizhugan decoction were 618.6μg·g-1and 49.09μg·g-1, respectively. Conclusion: The method is sensitive, rapid, efficient and easy to operate. It can be used for the determination of glycyrrhizin and protocatechuic acid in Lingguizhugan Decoction.
