摘要 目的:通过单因素实验优化大黄-3颗粒的水提工艺。方法:以饮片粉碎粒度、浸泡时间、液固比以及提取时间为影响因素,总蒽醌含量(包括芦荟大黄素、大黄酸、大黄素、大黄酚与大黄素甲醚)和干浸膏得率为评价指标,以单因素实验的方法优化大黄-3颗粒的提取工艺。结果:得出最佳条件为大黄-3粉末过10目筛,加16倍水,浸泡0.5 h,回流提取10 min。在此条件下总蒽醌含量为2.04 mg·g-1(芦荟大黄素0.33 mg·g-1、大黄酸0.35 mg·g-1、大黄素0.11 mg·g-1、大黄酚1.02 mg·g-1与大黄素甲醚0.23 mg·g-1),干浸膏得率为76.99%。结论:该方法稳定、合理、可行,可用于制备大黄-3颗粒。

[Abstract]
Objective: To optimize the water extraction process of Rhubarb-3 granules by single factor experiment. Methods: The extraction technology of Rhubarb-3 granules was optimized by single factor experiment, with the factors of granularity, soaking time, ratio of liquid to material and extraction time, the content of total anthraquinone (including aloe emodin, rhein, emodin, chrysophanol and physcion)and the yield of dry extract as evaluation indexes. Results: The optimum conditions were as follows: Rhubarb-3 powder was sieved through 10 meshes, added with 16 times water, soaked for 0.5h and refluxed for 10 minutes. Under these conditions, the total anthraquinone content was 2.04 mg·g-1(aloe emodin 0.33 mg·g-1, rhein 0.35 mg·g-1, emodin 0.11 mg·g-1, chrysophanol 1.02 mg·g-1 and physcion 0.23 mg·g-1), and the yield of dry extract was 76.99%. Conclusion: The method is stable, reasonable and feasible, and can be used to prepare Rhubarb-3 granules.
