目的:建立玫瑰花标准汤剂HPLC指纹图谱及含量测定方法,并与市售配方颗粒进行比较,为玫瑰花配方颗粒的质量控制提供参考依据。方法:采用同一HPLC方法建立玫瑰花标准汤剂指纹图谱及槲皮素与山柰素的含量测定方法,以十八烷基硅烷键合硅胶为填充剂;以甲醇-0.4%磷酸溶液(45∶55)为流动相;检测波长设为360nm,柱温设为30℃,流速为1.0ml/min。色谱数据采用国家药典委员会中药色谱指纹图谱相似度评价分析系统,生成标准指纹图谱。结果:槲皮素进样量在0.2489~24.8939 μg/mL呈良好的线性关系(r=0.9999),山柰素进样量在0.2135~21.3538 μg/mL呈良好的线性关系(r=0.9999),15批样品有5个共有峰,相似度为0.978~0.997,且与市售配方颗粒一致,通过与对照品比对指认出其中2个色谱峰。结论:建立的玫瑰花标准汤剂HPLC指纹图谱及含量测定方法,其特征性及专属性强,重现性良好,可作为玫瑰花标准汤剂及配方颗粒的质量控制方法。

Objective: To establish a HPLC fingerprint method and determination of Rosa rugosa Flos standard decoction, then compare with formula granules. To provide the basis for the quality identification of Rosa rugosa Flos standard decoction, Methods: The column was YMC-Pack ODS-A C18(250 mm×4.6 mm,5μm).The mobile phase was methanol-0.4% phosphoric acid (45:55),The detection wavelength was 360 nm.Column temperature was 30℃.The flow rate was 1.0 ml/min. The chromatographic fingerprint similarity evaluation software was used to analyze the data and evaluate the HPLC-fingerprints.Results: The linerar ranges of mcletin and kaempferide were 0.2489~24.8939 μg/mL(r=0.9999)and 0.2135~21.3538 μg/mL(r=0.9999). The HPLC fingerptint of 15 batches of Rosa rugosa Flos standard decoction was constructed with 5 common specific chromatogram peaks and 2 common peaks were identified by comparing with the reference standards. The similarity of 15 batches of Rosa rugosa Flos standard decoction was 0.978~0.997.Conclusion: The methed is species and reproducible. It can be used as the quality evaluation and species identification of Rosa rugosa Flos standard decoction.
