[摘要]
目的:探讨黄芩提取物对乳腺癌MCF-7细胞的生长抑制作用。方法:通过对经不同浓度黄芩提取物处理过乳腺癌MCF-7细胞染色,使用流式细胞仪进行细胞周期分析。结果:各组单因素方差分析结果显示,黄芩提取物作用于乳腺癌MCF-7细胞48h后10ug/ml和20ug/ml组细胞凋亡率均明显增加,与对照组比较均有差异(P<0.05);细胞周期检测结果显示,10ug/ml和20ug/ml组细胞G0/G1期明显增加,与对照组比较均有差异(P<0.05)。结论: 黄芩提取物可以诱导乳腺癌MCF-7细胞凋亡发生,从而在乳腺癌细胞中产生抗肿瘤的作用。

ABSTRACT Objective:Study the Effect of Baicalin on the apoptosisof Human Breast cancer cell line MCF-7. And explore the molecular mechanisms of Baicalin on the apoptosis.Method:Observe the changes of cell morphology after treatment of Baicalin by hematoxylin eosin staining.Use laser confocal microscope to observe the changes of fluorescence in MCF-7 cell cultured with Baicalin, by hoechest33342 staining.Result:HE staining shows the chromatin margination, karyorrhexis, apoptotic body could be observed in the MCF-7 cells cultured with Baicalin. While the MCF-7 cells in blank control grow well.According to laser confocal microscope, it could be found that the MCF-7 cell combined with Baicalin,have the same morphologic changes as HE staining. Furthermore, it also present line-like fluorescence in MCF-7 cell treat with Baicalin. These phenomena do not exist in blank control.Conclusion:Baicalin is a promising negative regulator for Human Breast cancer cell line MCF-7on proliferation.Baicalin could induce apoptosis of Human Breast cancer cell line MCF-7 and function as a Breast cancer treatment drug.
