摘要 目的 应用网络药理学方法分析半夏白术天麻汤治疗脑卒中的作用机制。方法 通过TCMSP、TCMID数据库获得半夏白术天麻汤各单味药成分并筛选出其治疗脑卒中的潜在靶点,应用网络药理学分析方法获得蛋白相互作用关系以及靶点参与的信号通路、生物过程,将蛋白相互作用关系导入Cytoscape软件并构建网络,CytoHubba插件筛选出关键靶点。应用AutoDock Tools、AutoDock Vina软件将得到的关键靶点与活性成分进行分子对接得到对接得分。 结果 通过数据库筛选得到98个半夏白术天麻汤治疗脑卒中的潜在靶点,KEGG富集通路筛选出靶点参与缺氧诱导因子1信号通路、肿瘤坏死因子信号通路等27条信号通路,GO功能富集分析结果表明,靶点参与了上皮细胞凋亡过程的负调控、血管重塑等163个生物过程。最终确定关键靶点为MAPK1、MAPK3、PIK3CA、PIK3CB、PIK3CD、mTOR、TNF、Bcl-2、IL-6。半夏白术天麻汤通过胫骨青霉素、8β-乙氧基苍术内酯Ⅲ、香风草甙、菜豆素、3β-乙酰氧基苍术酮、氢化松苓酸、异欧前胡素、甘草酚、反鳕油酸等成分达到治疗脑卒中的效果。 结论 半夏白术天麻汤能够通过多成分、多靶点、多通路治疗脑卒中,本研究为半夏白术天麻汤的临床应用提供了理论支持。

Abstract OBJECTIVE The mechanism of Banxia Baizhu Tianma Decoction in the treatment of stroke was analyzed by network pharmacology. METHODS Through TCMSP and TCMID databases, the single components of Banxia Baizhu Tianma Decoction were obtained, and its potential targets for the treatment of stroke were screened,The network pharmacological analysis method was used to obtain the protein interaction relationship and the signal pathways and biological processes involved by the target. The protein interaction relationship was introduced into Cytoscape software and the network was constructed. The cytohubba plug-in screened the key targets Using autodock tools and autodock Vina software, the obtained key targets were connected with the active components to obtain the docking score. RESULTS 98 potential targets of Banxia Baizhu Tianma Decoction in the treatment of stroke were screened through the database. The KEGG enrichment pathway screened 27 signal pathways involved in hypoxia inducible factor 1 signal pathway and tumor necrosis factor signal pathway,Go functional enrichment analysis showed that the target was involved in 163 biological processes such as negative regulation of epithelial cell apoptosis and vascular remodeling.Finally, the key target is MAPK1、MAPK3、PIK3CA、PIK3CB、PIK3CD、mTOR、TNF、Bcl-2、IL-6。Banxia Baizhu Tianma Decoctionhas has the effect of treating stroke through shinpterocarpin、8β- EtractylenolideⅢ、didymin、3βacetoxyatractylone、isoimeperaorin、trametenolic acid、isoimeperaorin、glycyrol、gadelaidicand other componentsCONCLUSION Banxia Baizhu Tianma decoction can treat stroke through multi-component, multi-target and multi-channel.
