[摘要]
目的:通过蛋白质组学分析对比研究山慈菇乙酸乙酯提取物(以下简称山慈菇酯提物)与阿霉素抗 4T1乳腺癌的作用靶点。方法:利用基于质谱的多肽体外标记技术( TMT),研究山慈菇酯提物/阿霉素影响4T1乳腺癌荷瘤小鼠的差异蛋白,并利用生物信息学技术解析不同细胞表达蛋白质的互作关系和生物功能。结果:蛋白质组学研究发现,以Fold change >1.5 或 < 0.67, P-value <0.05为筛选标准,山慈菇酯提物/阿霉素两种给药组抗乳腺癌的作用靶点相似,差异表达蛋白仅有13个,包括Ighg1、Ighm、Igkc、Gm5571及Cd5l等,这些差异表达蛋白主要与抗4T1乳腺癌免疫调节作用相关。其中Ighm、Igkc与Th1和Th2细胞因子分泌、Th17细胞分化、吞噬、细胞黏附有关。结论:山慈菇酯提物与阿霉素抗乳腺癌的作用靶点相似。

Objective:Proteomic analysis was carried out to comparative study effect target between ethyl acetate extract of Cremastra Appendiculata (Cr Ap) and Adriamycin (ADM) in Anti-4T1 breast cancer. Methods:In this study, mass spectrometr-based in vitro peptide labeling (TMT) technique was used to investigate the effect of Cr Ap/ADM on differential proteins in Anti-4T1 breast cancer Tumor bearing mice.Bioinformatics techniques were used to analyze the interactions and biological functions of the differentially expressed proteins. Results:Proteomics studies (Fold change >1.5 or < 0.67, P-value <0.05) showed that there were only 13 differentially expressed proteins, including Ighg1, Ighm, Igkc, Gm5571, Cd5l and so on. These differentially expressed proteins were mainly related to the immunomodulatory effect of Anti-4T1 breast cancer.Ighm and Igkc are related to Th1 and Th2 cytokine secretion, Th17 cell differentiation, phagocytosis and cell adhesion. Conclusion:The Anti-4T1 breast cancer targets were similar to that of Cr Ap and ADM.
