目的 探讨鹤草酚对前列腺癌细胞增殖、细胞周期和凋亡的影响。 方法 通过细胞计数、菌落形成试验、细胞周期试验、细胞凋亡试验等检测细胞增殖、细胞周期和细胞凋亡的变化。通过蛋白印迹法(western blotting)检测细胞凋亡相关蛋白的表达。通过小鼠异种移植瘤模型研究了鹤草酚在体内对肿瘤发生的影响。 结果 鹤草酚抑制了DU145细胞和PC-3细胞的增殖,降低了细胞存活率。与对照组相比,经鹤草酚处理后,S期DU145细胞和PC-3细胞的比例增加,而G2/M期的细胞比例降低。鹤草酚通过激活caspase 3相关的线粒体依赖性凋亡通路,促进DU145细胞和PC-3细胞凋亡。体内实验证实,鹤草酚能减缓前列腺癌细胞的形成,延长移植瘤小鼠的生存时间。 结论 鹤草酚是一种潜在抗癌药物,它通过激活凋亡通路,来抑制前列腺癌细胞的增殖和细胞周期进展。

[Abstract]
Objective: To investigate the effects of agrimophol on the proliferation, cell cycle and apoptosis of prostate cancer cells. Methods: The changes in cell proliferation, cell cycle and apoptosis were detected by cell counting, colony formation assay, cell cycle assay and cell apoptosis assay. The expression of apoptosis-related proteins was detected by western blotting. The effects of agrimophol on tumorigenesis in vivo were investigated by a mouse xenograft tumor model. Results: Agrimophol inhibited the proliferation of DU145 cells and PC-3 cells and decreased the rates of cell survival. Compared to the control group, the percentage of DU145 cells and PC-3 cells in S phase increased and the percentage of cells in G2/M phase decreased after treated with agrimophol. Agrimophol promoted the apoptosis of DU145 cells and PC-3 cells through the activation of caspase 3-related mitochondria-dependent apoptotic pathway. In vivo experiments confirmed that agrimophol slowed down the formation of prostate cancer cells and prolonged the survival of tumor-bearing mice. Conclusions: Agrimophol is a potential anticancer drug that inhibits the proliferation of prostate cancer cells and the progression of cell cycle by activating the apoptotic pathway.
