[摘要]
目的:利用网络药理学和分子对接技术研究文冠木活性成分的抗衰老机制。方法:基于TCMSP数据库及文献报道获取文冠木主要化学成分,将口服生物利用度(OB)、类药性(DL)设定为筛选参数,寻找活性成分,并通过Swiss Target Prediction数据库进行靶标预测;从TTD、GAD、PharmGkb数据库收集抗衰老的靶标;将活性成分靶标与抗衰老靶标取交集,在STRING数据库中将该交集绘制蛋白互作PPI网络,得到核心靶标;对核心靶标进行GO功能富集分析和KEGG通路富集分析;使用Cytoscape 构建“中药-活性成分-靶标-疾病-通路”网络得到核心成分,并对核心成分与核心靶标的结合模式进行分子对接验证。结果:获取金圣草黄素、槲皮素、山奈酚等29个化合物为文冠木抗衰老主要活性成分;筛选到AKT1、TNF、EGFR等30个基因为文冠木抗衰老的核心靶标。核心靶标主要涉及化学反应、离子束缚、胞质部分的活动等生物过程,通过参与PI3K-Akt 信号通路,脂质和动脉粥样硬化,癌症蛋白聚糖,丝裂原活化蛋白激酶信号通路,表皮生长因子受体酪氨酸激酶抑制剂抗性等来发挥抗衰老的作用。分子对接结果显示,排名前三的化合物与核心靶标AKT1、TNF、EGFR均能自由结合。结论:本研究证实文冠木的活性成分可通过多靶标、多通路发挥抗衰老作用,为未来深入研究文冠木在抗衰老和化妆品领域的作用提供科学依据。

[Abstract]
Objective: To explore the mechanism of active compounds in Xanthocerais lignum for anti-aging by means of network pharmacology and molecular docking.?Methods:Chemical compositions of Xanthocerais lignum were obtained by Traditional Chinese Medicine Systems Pharmacology (TCMSP) and literature reports, Oral bioavailability (OB) and drug-like properties (DL) were set as screening parameters to search for active ingredients;Swiss Target Prediction database were used to predict the active component; Anti-aging targets were collected from TTD,GAD and PharmGkb databases;?PPI protein interaction network was constructed through STRING database, and core targets were screened; Enrichment analysis of gene ontology (GO) function and kyoto encyclopedia of genes and genomes (KEGG) pathway were performed to explore the possible biological functions and pathways of core targets.?Cytoscape software was used to construct a visual network diagram of "TCM - active component - target - disease - pathway" to obtain the core components, and the binding mode of the core components and targets were verified by molecular docking.?Results:Twenty-nine components,including chrysoeriol,quercetin,kaempferol,were obtained as the main anti-aging components of Xanthocerais lignum.?Thirty genes including AKT1,TNF and EGFR were screened as the core targets of anti-aging.?Core targets mainly involved in chemical reactions,ionic bonds,cytoplasmic part of the activities of the biological process,by participating in pathways such as PI3K-Akt signaling pathways,lipid and atherosclerosis,proteoglycans in cancer,MAPK signaling pathway to play the role in anti-aging.?Molecular docking showed that the top three compounds could freely bind to the core targets AKT1, TNF and EGFR. Conclusion:This study preliminarily confirmed that the active components of Xanthocerais lignum could exert anti-aging effects through multi-target and multi-pathway,and provided scientific basis for further research on the anti-aging effects of Xanthocerais lignum.
