摘 要:目的 采用网络药理学探索蒙药肉豆蔻治疗缺血性心脏病的作用机制。方法 课题组主要通过中药系统药理学分析平台(TCMSP),检索得到蒙药肉豆蔻的主要化学成分以及作用的靶点。使用Uniprot数据库,检索与靶蛋白相对应的基因,运用软件(Cyoscape 3.7.2)绘制“化合物-靶点-通路”网络模型进行可视化,通过DAVID数据库进行GO分析以及KEGG通路分析,预测其作用机制。结果 “化合物-靶点-通路”网络模型中包含10个化合物以及与之对应的靶点54个,关键性靶点涉及PTGS2、KDR、MMP9、ESR1、JAK2等。肉豆蔻治疗缺血性心脏病涉及催乳素信号通路、雌激素信号通路、炎症信号通路、癌症通路等多条信号通路。结论 本文将网络药理学运用于蒙药肉豆蔻治疗缺血性心脏病中,筛选活性成分、靶点蛋白、相关信号通路,将三者结合绘制“活性成分-靶蛋白-通路”网络图并预测其作用机制,为后期研究蒙药肉豆蔻治疗缺血性心脏病提供新的研究思路。

[Abstract] Objective To explore the Mongolian medicine nutmeg in the treatment of ischemic heart disease by network pharmacology. Methods The research group mainly used the Traditional Chinese Medicine System Pharmacology Analysis Platform (TCMSP). The gene corresponding to the target was queried through Uniprot database, and then the "compound-target-pathway" network was constructed by Cyoscape 3.7.2 software for visualization. DAVID was used for GO function enrichment analysis and KEGG pathway enrichment analysis to predict its mechanism of action. Results the "compound-target-pathway" network includes 10 compounds and 54 corresponding targets. the key targets include PTGS2, KDR, MMP9, ESR1, JAK2, etc. Nutmeg treatment of ischemic heart disease involves prolactin signal pathway, estrogen signal pathway, inflammation signal pathway, cancer pathway and other signal pathways. Conclusion In this paper, network pharmacology is applied to Mongolian medicine nutmeg in the treatment of ischemic heart disease. Active components, target proteins and related signal pathways are screened, a "compound-target-pathway" network diagram is constructed and its mechanism of action is predicted, which provides new research ideas for later research on Mongolian medicine nutmeg in the treatment of ischemic heart disease.
