目的 尿激酶型纤溶酶原激活因子是一种新型的肿瘤生物标记物,本课题就是研究PLAU在头颈部鳞状细胞癌中的表达以及转移中的作用。方法 将从中科院细胞库购入的癌细胞送至基因实验室,进行基因测序及富集分析,并采用qPCR、Western-blot实验、细胞划痕实验以及Transwell实验技术分析肿瘤细胞的迁移能力,检测N-钙粘蛋白、锌指转录蛋白、波形蛋白在HNSCC细胞中表达的情况分析其表达与PLAU对肿瘤远处转移的相关性。结果 PLAU在HNSCC中高表达(p<0.05),且晚期或复发转移性HNSCC中的PLAU明显高表达于早期或原位癌。过表达PLAU有促进喉癌细胞转移的能力,PLAU可能调控N-钙粘蛋白、锌指转录蛋白、波形蛋白等诱导头颈部肿瘤的转移(p<0.05)。结论 PLAU可能是通过细胞-间充质转化过程促进HNSCC的迁移和侵袭。

Objective Urokinase type plasmin is a new type of tumor biomarker. This topic is to study the expression of PLAU in head and neck squamous cell carcinoma and its role in metastasis. Methods Send cancer cells purchased from the Chinese Academy of Sciences cell bank to the gene laboratory for gene sequencing and enrichment analysis. The migration ability of tumor cells was analyzed by qPCR, Western blot test, cell scratch test and Transwell test. The expression of N-cadherin, zinc finger transducer, Vimentin in HNSCC cells was detected to analyze the correlation between their expression and PLAU's effect on tumor metastasis. Result PLAU is highly expressed in HNSCC (p<0.05), and PLAU is significantly overexpressed in advanced or recurrent metastatic HNSCC compared to early or in situ cancer. The overexpression of PLAU has the ability to promote the metastasis of laryngeal cancer cells. PLAU may regulate the metastasis of head and neck tumors induced by N-cadherin, zinc finger transcription protein , Vimentin, etc. (p<0.05). Conclusion PLAU may promote the migration and invasion of HNSCC induced cell mesenchymal transition.
